With the growth of Twitter windows Twitter clients are becoming very important. If you don't manage your followers properly Twitter can become very time consuming and end up being a big time waster. Tweeting from web is alright if you have a small following but if you have a considerable following then you can easily miss important tweets that way. This is one feature that was missing in TweetDeck that almost made me leave TweetDeck. But the latest version had multiple account support so now I have no reason to leave it.

If you are following a considerable amount of people your timeline can get cluttered pretty quickly. This can lead to you missing important tweets. Now you can group your followers according to your preference. This enables you to easily track tweets of different users.
Multiple columns
The perfect feature to compliment the grouping feature. You can have a different column for different groups and you can have separate columns for replies direct messages etc
Facebook Friend Status updates
Now you don't have to be logged into Facebook to view your friends status. You can view them through TweetDeck and you can update your status using TweetDeck as well.
Url shortening services available through many services
Add the url you want to shorten to the shorten url text box and shorten the url using one of the many services available , no need to visit another site
Synchronize across multiple computers
Now you can set up a column or group from your home and view them from your office. You need an account for this but definitely worth it if your tweeting from both locations.
TwitPic support
TwitPic is one of the most used Twitter application around to upload pictures to Twitter and that service is integrated to TweetDeck. You just have to click the TwitPic button and browse for the picture to upload.
Support for StockTwits
This might not be useful for everybody , but if you are into stock market and trading then this is a great addition to the tool
